It would make a really great story but 40k is much to cemented to throw something in like that. That's just cheap writing to get attention. There'd be little point in such a thing.
Destroyed, yes, but why and by whom? No records exist so GW may be able to write lore on the fallen legion. Did the Emperor save the gene seed? Yes, Greg Knights were not formed until later, but that does not explain away that their geen seed may not be from a missing Primarch. Anywhere I have looked for this information has been vague at best, which seems to be by design.
Vaults of Terra suggested that GK gene seed probably came directly from the emperor as there was no primarch gene seed available due to all of that heresy stuff that was going on at the time.

Theres not much to talk about really, unless they add new factions (IG, SoBs and their chaos counterparts) The only faction with a gender option would be the eldar. Of course if the devs actually decide to invest time on it, which may be unlikely.
